2. Leukocyte Classification by Flow Cytometry
This method differentiates blood cells on the basis of the scattered light and fluorescent light generated by irradiating the cells passing through a flowcell with a laser beam. The scattered light contains information mainly on the size of the cell, shape of nucleus and the presence of granules. The fluorescent light is generated by staining blood cells with a special fluorescent dye, and contains information mainly on the nucleic acid (DNA and RNA) content of the blood cell. In 1998, Sysmex developed the R-1000, which used flow cytometry and was the world's first automated reticulocyte analyzer. This method has been adopted in 5-differential WBC analyzers, including those of the XE series.
